<p> As a first step to contribute to this year's iGEM competition, we decided to participate in iGEM's fifth International Interlaboratory Study along with many other teams around the world. This study is organized by iGEM's measurement committee in an effort to establish standardized, reliable and repeatable measurement tools for the iGEM community and the synthetic biology community as a whole. This year's iGEM InterLab study is about establishing a standardized protocol for the measurement of GFP using a plate reader. To start things off, we needed a plate reader, Tecan Infinite M200 Pro plate reader, which is qualified to measure GFP fluorescence. Additionally, we needed competent E.coli cells DH5α. These were prepared from glycerol stocks. Together with the material iGEM had provided we were ready for work. Throughout our experiments, we tested 8 plasmids (2 controls and 6 test devices) by measuring the OD600 and the fluorescence of the cells carrying the plasmids. Besides, we calculated the Colony Forming Units per 0.1 OD600 E. coli cultures. The workflow can be separated into six segments.  </p>
