Biosafty

  Overview

According to different scenarios, we designed different promoters to combine with mazF. With the humanization design, the biosafty of our projects is guaranteed.

    Bromidrosis

We introduced a mazF suicide system into E. coli. This system is induced by arabinose. We designed a circuit to verify the function of this suicide system, which was araC+pBAD+RBS+mazF. We constructed a plasmid containing the circuit, and transformed it into E.coli BW25113 respectively. We verified the effect of different arabinose concentrations on the lethal effect of mazF. We used E. coli without added arabinose as a Negative control and used 5 different concentrations of arabinose to induce the mazF suicide switch (5%, 10%, 15%, 20%, 50%). We add arabinose 5 uL into 5 mL LB culture medium when the OD600 is 0.6 (the log phase). We observed the survival of E. coli every hour.

mazEF

According to the experimental results, we can know that mazF has a strong lethal effect. The death rate of E.coli connected with the concentration of arabinose. In the range of commonly used arabinose concentrations (5% to 50%), the higher the concentration, the more the amount of mazF that is induced to be expressed, resulting in more death of E. coli. Two hours after the addition of arabinose, E. coli almost all died. Therefore, we decided to use 50% arabinose in the future scent spray to induce the mazF suicide switch, which will make our kill switch work more efficiently.

    Garbage can

In the trash can lid, we chose a light-controlled promoter to combine with mazF. Once the engineered bacteria leaves the dark environment of the trash can, it will be killed.
