<p>Project goal: to genetically modify a bacteriophage capable of supressing toxin production in C. difficile by integrating our genetic constructs into its genome. We are using a bacteriophage known to infect strains of C. difficile called phiSBRC. phiSBRC will be packaged with constructs to supress toxin production either via a dead-Cas9 approach or via antisense RNA (asRNA).

When creating the antisense RNA parts, we decided to aim for 2 well characterised toxins in C. difficile, TcdA and TcdB. The parts were constructed in such a way that they targeted both toxins simultaneously because both toxins have been shown to have independent roles.<p>
