iGEM Winter Camp

Why: To pass on the spirit of iGEM as well as the passion and love for synthetic biology
How: A 5-day winter camp for the next young generation in our school What:
After a long period of preparation, we held a winter camp for the junior high school students at grade 9 beginning on February 3rd. During this camp, we made a brief introduction about synthetic biology to them. After that, we taught them how to use pipette--one of the most important tools in synthetic biology and entertaining them by doing an interesting experiment of DNA extraction via bananas. In addition, we taught them some basic experiment techniques that would surely come in handy in the future, including plasmid extraction, PCR, transformation, and running agarose gel. Meanwhile, we scatter around to help with their operation. Reflection:
Although feeling exhausted after 5-day hard work, we did feel a strong sense of accomplishment. So diligent were they that they even discussed the principles and methods of an experiment with us during break time! Young as we are; however, we had almost organized the whole camp and acted as the lecturer, and that wasn’t an easy task, though. With this experience, we equipped with the capability of teaching others of what we’ve known and will never panic while dealing with any hard task in the future. Course Review:
1. Building with Biology We discussed the pros and cons of GMO foods with the participants, and did a rough survey on this issue. We then did Banana DNA extraction using everyday items such as dishwasher, salt, and hot water to show that DNA isn’t the stuff of myths, but exists all around us. 2. Pipette Handling and Usage Team members showed the participants how to handle a pipette, and taught them the basics of using lab equipment. 3. Plasmid Extraction To extend the banana DNA extraction experiment, we taught the participants how to extract plasmid DNA from bacteria 4. PCR The participants were taught the main principles of PCR (Denaturation, Annealing, Extension and Amplify). Team Mingdao used a short film to explain the long process, then we let the participants to try on themselves. 5. Restriction Enzyme The principle and the application of restriction enzymes were presented. Our PI then taught the participants about compatible ends of biobrick parts. 6. Transformation Heat-shock method and bacterial culture with antibiotics were emphasized in this section. 7. DNA Agarose Gel Electrophoresis We used the blue LED viewer to see the results of PCR and DNA extraction done the day before. 8. Biolab tour The participants are given a throughout tour of the lab and taught the lab rules.
