SifA maintains the integrity of Salmonella-containing vacuole (SCV) where Salmonella survive and replicate1. The exsistence of SCV limits GSDMD-N275 releasing to cytoplasma. what else, growth of ΔsifA mutant is inhibited in macrophage2.Thus, we knock out the sifA gene in order to prevent the stability of SCV and reduce the virulence of Salmonella. ΔsifA mutant was cnstructed by using gene editing systerm based on two-step allelic exchange3.
