Tell us about your project, describe what moves you and why this is something important for your team.

We have been working on the plasmid design and I think we are set to synthesize a couple parts from IDT. We will synthesize the SaCas9 and the CRISPR testing platform containing the target sequence and EricaCFP. The SaCas9DNA contains the LacI promoter and gene. We will then insert the gRNAtracerRNA into SaCas9 and SpCas9 at EcoNI and SgrAI sites. We will then clone in the SaCas9 into one of the backbones that Igem provides us with EcoRI and PstI. Before we clone in the SpCas9 into a different backbone, we must PCR in restriction sites EcoRI (left of LacI) and PstI (Right of Cas9) to be able to clone into the backbone. The pET-Cas9 DNA included contains all of the primers that we will be working with for PCR, we have to take out XbaI and EcoRI because of Igem requirements. We will then clone in the SpCas9 into a different Cas9 and continue with the point mutations that lead to dCas9 and continue with experiments.
