Bioremediation of Sour Crude Oil Waste using Cyanobacteria




Safety of the project

Our project used two non-pathogenic strains of E.coli: DH5alpha and Top10. Those strains were used for transformation and amplification of plasmids pSyn_6 with ligated SQR. They were also used for amplification of pSB1C3 plasmid and gene submission. Miraprep protocol was used for DNA plasmid extraction.

The second organism used in the project is cyanobacterial organism Synechococcus elongatus PCC 7942 strain. This is a transformable strain of freshwater non-pathogenic model organism used for investigation of molecular genetics.

Mandatory Rules of Safety include:
  • Gloves, coat, closed shoes and tied up hair
  • Glasses in the case of working with irritant substances
  • Washing hands with detergents before and after lab work
  • Sterilization of workplace with 70% ethanol
  • Disposal of organisms into organic wastes after 10% bleach treatment
  • Work in the fume hoods in case of contact with volatile and harmful reagents
  • Safety cabinet BSL1 for sterile work with bacterial and cyanobacterial cultures
