Besides our 3G parts library, we are also submitting a series of heat inducible reporters (mScarlet) and proteases (mf-Lon), which we are using to construct IFFLs with different parameters. We have varied the promoter strength through the use of either J23107 or J23100. For a repressor we used either temperature sensitive lambda-cl which induces at 37 C, or TlpA39, which induces at 39 C. To alter degradation rate we used ptds, protein degradation tags, of varying strengths, or ssrA, another type of degradation tag. By combining these parts in various combinations, we were able to test IFFLs of various strengths and parameters.
