With the use of site-direted mutagenesis the goal was to improve the part of last years LiU iGEM team biobrick BBa_K2474000 and make it to a reporter system which could be more widely used. The reason we choose to improve this part was not only to create a functional reporter system, but also to illustrate the loss of exchange with a aggregation prone fusion tag. From fig 2. you can clearly see that the version without amyloid-beta 1-42 folds better and thus gives a much higher fluorescence, while the fusion version is having trouble.
