Description

With the growth of synthetic biology, there has been an increase in the development of digital synthetic circuits. They require biological logic gates that can accept a binary input and generates a suitable output. But biological systems may not always exist at two stable concentrations. Hence there exists a need of robust and reliable circuits that can transform the analog and stochastic behaviours of biology into a digital response. Our elementary constructs can then be used to develop complex circuits with specialized functions.

Recombinases are enzymes that have proven to be highly robust and efficient. They trigger site-specific recombination to perform excision/incision or inversion of genetic circuits, exclusively or in tandem to produce the desired gene expression. Our project involves use of serine based recombinases to develop a novel recombinase based toolkit of elementary circuits such as switch, feedforward loop, feedback loop etc. This toolkit will allow researchers to develop modular and orthogonal systems that can be tuned provide the desired output.
