After producing our target protein---bacteriocin, we than want to check whether these bacteriocins inhibit the growth of Bacillus subtilis or not. Therefore, we test the inhibition ability of bacteriocin to know each bacteriocin’s Minimum Inhibitory Concentration (MIC). Our MIC Test will be separated into two parts. One is Inhibition Zone, and the other is Microdilution.

Inhibition Zone
To test the inhibition ability of bacteriocin, we use Inhibition Zone to measure the effective inhibition range under different dose of bacteriocin.
Microdilution
We use Elisa Reader to record OD600 values of samples once an hour, if OD values decrease, it means that our bacteriocin inhibit the growth of Bacillus subtilis. Further, we can also know when does bacteriocin degrade by observing the growth curve of Bacillus subtilis.
