As we all know, smoking is extremely harmful to health, and secondhand smoke is very easy to cause damage to public health. In daily life, people have been suffering a lot.

Nicotine, which stems from ignited cigarettes, is so addictive that it is a sort of aromatic poison to smokers.

Through literature reviewing, it is found that nicotine has adverse effects on many tissues or organs of human body and is a major threat to human health.

At the same time, the high content of nicotine in tobacco-leave waste will pose a serious threat to the safety of groundwater in China.

Scholars at home and abroad have been paying attention to the degradation of nicotine by microorganisms. Comparing with physical and chemical methods, microbial degradation of nicotine is more economical, more efficient and more sustainable.

Abstract


Therefore, we hope to provide a simple and low-cost biological monitoring tool of nicotine content for the public and relevant environmental protection departments.

We have designed a portable blow-type biological detector based on Escherichia coli, which can be used to detect dissolved nicotine in water and approximate nicotine content in air.

At the same time, we constructed several plasmids that could specifically respond to the presence of nicotine, and confirmed that two of them could work specifically.

CONTAINING:

The first generation of the plasmids: the nicotine induced NicA2 promoter + the reporter gene GFP;

The second generation of the plasmids (The aim of constructing the second generation is to enhance the expression level of GFP): the nicotine induced NicA2 promoter + the reporter gene GFP;

And a generation of failed plasmids: possible nicotine induced Nox promoter (which was later confirmed to have no specific expression of the presence of nicotine) + the reporter gene BFP and possible nicotine induced Nox promoter + the reporter gene RFP

All in all, we are struggling to remove the secondhand smoke public and make sure there is no threat to public health as much as possible.
